“Bioresorbable, electrospun nonwoven fabric as a multidrug delivery system for combined therapy of glioblastoma multiforme”

The aim of the project is to develop a multidrug delivery system in the form of a nonwoven fabric for controlled release of an anticancer drug, an autophagy inhibitor and a radiosensitizer, as a proposal for a future, combined with radiation, method of treating glioblastoma multiforme.
The designed DDS will be based on bioresorbable polymers and obtained by electrospinning and electrospraying. Three drugs with different mechanisms of action were selected for the study in order to enhance the anticancer effect. The prolonged but delayed drug release will be optimized to correlate the start of chemotherapy with irradiation – core-shell and mixed fibers will be used, but also the appropriate order of nonwoven layers. The results will help to understand the kinetics and mechanisms of drug release from bioresorbable nonwovens, which in turn are related to the degradation mechanism of the polymer carrier, but will also allow to predict and adjust the doses of delivered drugs, allowing the adaptation of polymers to specific biomedical indications. The efficacy of the proposed nonwoven will be verified in vitro and in vivo in an animal model.
